It will be automatic after that; TLU will install GPG from the TeX Live Manager developer's website, if you don't already have it, and verification will be transparent from then on (or until you install a new version of MacTeX). You'll only be aware of it if it fails.

Not that I recall, but you can check the log window to see for sure; just don't pay too much attention to it, as 99% of that is only useful to me for debugging the program. Whenever you launch TLU it should have something about GPG printed to the log output.
