This depends on what you intend to do! /usr/local/texlive/texmf-local is this system's tree of local extensions, additions, corrections, set-ups which is valid for all users of this system. Its contents has to be "hashed", i.e., 'mktexlsr /usr/local/texlive/texmf-local' has to be run with elevated privileges, i.e., with a sudo in front of the command.
~/Library/texmf is your own private ground. No mktexlsr/texhash, no sudo needed. Your private ground comes first when a TeX related commands searches for installed files. Then follows the system's local extension. Then comes the TeX distribution.
