Howdy,

That comment line is read by TeXworks (and will work with the latest versions of TeXShop on a Mac too). It tells TeXworks to use an IniiXeTeX Tool (or engine in TeXShop parlance) to compile the file. For this to work you need to define such a Tool (engine) since that isn't defined in the default setup. Most likely it simply runs

xetex -etex -ini 

on the file you are compiling.
