No.  It's supposed to work invoked from anywhere, like any normal
program.  I repeat: do binaries built by other people (macosx, w32,
cygwin, alpha-linux, i386-solaris) exhibit the same problem?

    i don't know how it is supposed to find its runtime files in the
    texmf tree.

Bowman implemented alternative code that runs kpsewhich to find its
files based on SELFAUTOPARENT.  You have to say --enable-texlive-build
when you configure it (see build.html#asymptote).  Did you do that?
