Hans Hagen has a way to get spot colours using Context, and Metapost I 
think it is---text coloured so though winds up in a .pdf as a ``form'' 
object which is resistant to standard pdf preflight techniques though.

You can carry my technique of placing each character in a .eps and 
stitching them together in a virtual font a step further and create a 
duplicate font where each .eps is set to be a particular spot colour 
though.

For my part, I've always either post-processed the .pdf or PostScript 
file, or just used CMYK, leaving out magenta and yellow, and telling 
the printer at press time to suppress those plates / films and to use 
Cyan in lieu of (say) Pantone 301.
