There were some changes recently, did you reinstall?

I think we agreed that we cannot make it work out of the box and
that users have to take some actions. But I thought that the
default setting in texmfcnf.lua is still shipped as TEXMFVAR for the
luatex cache. So the cache should be created in ~/.texlive2009/temxf-var
if I am not wrong?

Herbert: Do you have 
	ROOT/texmf/web2c/texmfcnf.lua
which basically says (with some comments and braces):
	TEXMFCACHE = '~/.texlive2010/texmf-var'
?
