That should run. I assume you have other tools (notably TeX) installed 
through i-Installer?
Can you post the output of echo $PATH
Can you post the output of file /usr/local/bin/bibtool

To my surprise bibtool does not respond to -h or --help (I thought that 
that was one of the good conventions brought about by GNU), but man 
bibtool gives a description on how to use it. Without input files, it 
expects input from the terminal, so you'd have to type the entries by 
hand if you just run the program (exit it with ctrl-C or ctrl-D).
