The problem with spotcolor.sty is that it requires the pdftex driver for the graphics package --- which doesn't work in XeLaTeX.<br>I tried to change spotcolor.sty to load the xetex driver, but I got a clash option.<br>

You can post-process the .pdf afterwards using Enfocus PitStop to assign the required spot colours, or use the old printing fallback of 100% cyan == spot colour FOO.<br>

If it's only regular repeating elements / reversed out text which need to be coloured you could prepare such elements in advance using a tool which supports spot colours, then place those using XeTeX, then overprint the solid area w/ white XeTeX text to knock it out.<br>
