As Herb mentioned: you need to download and install the "experimental" microtype package from the TLcontrib site. Tlmgr on all platforms and TLU (TeX Live Utility) on modern Mac OS X platforms (Snow Leopard, Lion) can perform this task completely. (The package is installed in the usual tree, not the local – or the personal – one, so you need to perform this task in TeX Live 2010 and again in TeX Live 2011.)
