Before fiddling about with CFLAGS and LDFLAGS it might be worth
to look at the option --with-kpathsea of the configure script.
E.g.

 --with-kpathsea=/usr/local/TL2007/lib

Other frequently needed options concern freetype2's include/lib paths:

 --with-ft2include=/usr/include/freetype2

etc.
